Product Overview
Clone 18S60 is a rabbit monoclonal antibody that recognizes splicing factor 3B subunit 1 (gene SF3B1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 145.8 kDa, and the supplier reports an observed band at about 155 kDa.
Splicing factor 3B subunit 1 (SF3B1, SF3b155) is the largest subunit of the SF3b complex in the U2 snRNP. It contacts the pre-mRNA around the branch site and helps position the branch-point adenosine, and recurrent SF3B1 mutations in myelodysplastic syndromes and other cancers, as well as splicing-modulator drugs that bind SF3b, make it a major research target.

Samples with a confirmed band (WB): human HepG2 cells, human HeLa cells, human RT4 cells, rat brain tissue, rat C6 cells, mouse brain tissue, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colon tissue, human liver tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human SF3B1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places splicing factor 3B subunit 1 in the nucleus and nucleus speckle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
