Product Overview
This rabbit polyclonal antibody targets serine/threonine-protein kinase Sgk1 in human and monkey samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 48.9 kDa, and the supplier reports an observed band at about 55 kDa.
Serum- and glucocorticoid-regulated kinase 1 (SGK1) is a serine/threonine kinase of the AGC family whose expression is rapidly induced by hormones and cell stress. Activated downstream of PI3K through mTORC2 and PDK1, it regulates many ion channels and transporters, notably the epithelial sodium channel in the kidney, and also affects cell survival and proliferation.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Monkey) |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL (Human) |
| Immunocytochemistry / Immunofluorescence | 2 µg/mL (Human) |

Samples with a confirmed band (WB): human HeLa cells, monkey kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:100, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human rectal cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): SiHa cells.
Immunogen
Synthetic peptide from the N-terminal region of human SGK1. Sequence identity with the rat ortholog: 95.7%.
Reactivity Notes
The supplier lists reactivity with human and monkey. UniProt places serine/threonine-protein kinase Sgk1 in the cytoplasm, nucleus, endoplasmic reticulum membrane and cell membrane. Tissue expression noted by UniProt (human): Expressed in most tissues with highest levels in the pancreas, followed by placenta, kidney and lung. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
