Product Overview
This rabbit polyclonal antibody targets SH3 and multiple ankyrin repeat domains protein 3 (gene SHANK3) in human, mouse and rat samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 184.7 kDa, and the supplier reports an observed band at about 180–190 kDa.
SHANK3 is a major scaffold protein of the postsynaptic density that organizes glutamate receptor complexes, via GKAP/PSD-95 and Homer, and links them to actin. It is critical for dendritic spine formation and synapse maturation, and SHANK3 loss causes Phelan–McDermid syndrome and is a well-known autism risk gene.

Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human meningoma tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): Raji cells, HEPA1–6 cells, NRK cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places SH3 and multiple ankyrin repeat domains protein 3 in the cytoplasm, postsynaptic density and cell projection. Tissue expression noted by UniProt (human): Expressed in the cerebral cortex and the cerebellum. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
