Product Overview
This rabbit polyclonal antibody detects tyrosine-protein phosphatase non-receptor type 6 (gene PTPN6) in human and mouse samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 67.6 kDa, and the supplier reports an observed band at about 68 kDa.
SHP-1 (PTPN6) is an SH2-domain tyrosine phosphatase expressed mainly in blood cells. Recruited to inhibitory receptors that carry ITIM motifs, it dephosphorylates signaling proteins to dampen immune cell activation and growth factor signaling, and it is a key negative regulator studied in immunology and hematopoiesis.

Samples with a confirmed band (WB): human Jurkat cells, human Raji cells, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with a confirmed signal (flow cytometry): A431 cells.

Recombinant human SHP1/PTPN6 fragment (Glu67–Lys572), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places tyrosine-protein phosphatase non-receptor type 6 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Isoform 1 is expressed in hematopoietic cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
