Product Overview

Clone GCA-16 is a rabbit monoclonal antibody that recognizes tyrosine-protein phosphatase non-receptor type 11 (gene PTPN11) in human samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 68.0 kDa, and the supplier reports an observed band at about 70 kDa.

SHP2 (PTPN11) is a cytoplasmic tyrosine phosphatase with two SH2 domains that, unusually, promotes signaling downstream of receptor tyrosine kinases and cytokine receptors, especially activation of the RAS–MAPK pathway. Germline PTPN11 mutations cause Noonan syndrome, and SHP2 is an active target in cancer drug research.

Validated Applications

Western blot 1:1000–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:50

Samples with confirmed staining (IHC): human heart tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human SHP2.

Reactivity Notes

The supplier lists reactivity with human. UniProt places tyrosine-protein phosphatase non-receptor type 11 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Widely expressed, with highest levels in heart, brain, and skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
