Product Overview
This rabbit polyclonal antibody targets sialate O-acetylesterase (gene SIAE) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 58.3 kDa, and the supplier reports an observed band at about 70 kDa.
Sialate O-acetylesterase (SIAE) removes 9-O-acetyl groups from sialic acids, acting on free sialic acid in the cytosol and on sialylated glycoconjugates in lysosomes. Working opposite sialate O-acetyltransferases, it sets the level of O-acetylated sialic acid on glycans, a modification that influences cell recognition, B-cell tolerance and interactions with pathogens.

Samples with a confirmed band (WB): human RT4 cells, human U251 whole cel, rat testis tissue, rat liver tissue, rat RH35 cells, mouse testis tissue, mouse liver tissue, mouse HEPA1–6 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human glioblastoma tissue, human thyroid papillary carcinoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): Caco-2 cells.
Samples with a confirmed signal (flow cytometry): HepG2 cells.
Immunogen
Recombinant human SIAE fragment (Arg230–Lys523), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Widely expressed with high expression in the testis, prostate, and colon. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
