Product Overview
Clone GG-19 is a rabbit monoclonal antibody that recognizes NAD-dependent protein deacetylase sirtuin-1 (gene SIRT1) in human samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 81.7 kDa, and the supplier reports an observed band at about 120 kDa.
Sirtuin 1 (SIRT1) is an NAD+-dependent deacetylase found mainly in the nucleus. It removes acetyl groups from histones and from many transcription factors and cofactors, including p53, FOXO proteins and PGC-1α, linking the cell's energy status to gene expression, DNA repair and survival, and it is widely studied in metabolism, stress responses and aging.

Samples with a confirmed band (WB): human Jurkat cells, human HeLa cells, human 293T cells, human K562 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human kidney tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Immunogen
Synthetic peptide based on the sequence of human SIRT1.
Reactivity Notes
The supplier lists reactivity with human. UniProt places NAD-dependent protein deacetylase sirtuin-1 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
