Key elements and design rationale
- Antibody format: Rabbit Polyclonal Rabbit IgG
- Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of mouse SLC10A1, different from the related human sequence by eighteen amino acids, and from the related rat sequence by three amino acids.
- Molecular weight context: reported MW: 50 kDa; calculated MW: 39413 MW
- Reactivity: Mouse,Rat
- Applications: Flow Cytometry, IF, IHC, IHC-F, WB
As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.
Biological background
Sodium/bile acid cotransporter; Sodium/bile acid cotransporter. Na+-taurocholate cotransporting polypeptide (NTCP), also known as SLC10A1 (Solute carrier family 10, member 1), is the major bile acid uptake system in human hepatocytes. NTCP and the ileal transporter ASBT (apical sodium-dependent bile acid transporter) are two sodium-dependent transporters critical for the enterohepatic circulation of bile acids. The hASBT gene is known to be activated by the glucocorticoid receptor (GR). Ho RH et al. indicates functionally important polymorphisms in NTCP exist and that the like lihood of being carriers of such polymorphisms is dependent on ethnicity. Functional note: The hepatic sodium/bile acid uptake system exhibits broad substrate specificity and transports various non-bile acid organic compounds as well. It is strictly dependent on the extracellular presence of sodium. Reported localization: Membrane; Multi-pass membrane protein. Expression/tissue context: Predominantly expressed in the brain.
Research relevance and current trends

- Plasma Membrane: Researchers commonly examine how Slc10a1 (Sodium/bile acid cotransporter) relates to this theme using model systems and orthogonal readouts.
- Signal Transduction: Researchers commonly examine how Slc10a1 (Sodium/bile acid cotransporter) relates to this theme using model systems and orthogonal readouts.
Common research applications
- Western blotting: compare relative Slc10a1 (Sodium/bile acid cotransporter) levels across conditions; band patterns may reflect isoforms and processing.
- IHC/IHC-F: assess spatial distribution of Slc10a1 (Sodium/bile acid cotransporter) across tissue regions and cell types using matched controls.
- IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
- Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.
Notes for experimental interpretation
- Specificity notes: No cross reactivity with other proteins.
- Cross-reactivity: No cross-reactivity with other proteins.
- Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
- Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
