Product Overview
This rabbit polyclonal antibody recognizes sodium- and chloride-dependent transporter XTRP3 (gene SLC6A20) in human, mouse and rat samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 65.9 kDa, and the supplier reports an observed band at about 70 kDa.
SIT1 (SLC6A20, XTRP3) is a sodium- and chloride-dependent transporter for imino acids such as proline and for N-methylated amino acids, and also carries glycine. In the brain it helps regulate proline and glycine levels and can thereby influence NMDA receptor activity.

Validated Applications
| Western blot | 0.1–0.25 µg/mL (Human, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |

Samples with a confirmed band (WB): human CCRF-CEM cells, human COLO 320 cells, human SW620 cells, human 293T cells, rat kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colon tissue, human liver cancer tissue, human lung adenocarcinoma tissue, human ovarian cancer tissue, human rectum adenocarcinoma tissue, mouse colon tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): 293T cells.
Immunogen
Recombinant human SLC6A20 fragment (Ser124–Ala555), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 91.9% and 90.3%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places sodium- and chloride-dependent transporter XTRP3 in the apical cell membrane. Tissue expression noted by UniProt (human): Kidney and small intestine. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
