Product Overview
Clone FGG-4 is a rabbit monoclonal antibody that recognizes diablo IAP-binding mitochondrial protein in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 27.1 kDa, and the supplier reports an observed band at about 21 kDa.
DIABLO (also called Smac) is a mitochondrial protein released into the cytosol during apoptosis. It binds inhibitor of apoptosis proteins such as XIAP and blocks their ability to restrain caspases, thereby promoting caspase activation downstream of cytochrome c. Smac mimetic compounds are based on its IAP-binding motif, and DIABLO is studied in apoptosis research.

Samples with a confirmed band (WB): Jurkat cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Samples with confirmed staining (IHC): human kidney tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Immunogen
Synthetic peptide based on the sequence of human smac/Diablo.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places diablo IAP-binding mitochondrial protein in the mitochondrion and cytoplasm. Tissue expression noted by UniProt (human): Ubiquitously expressed with highest expression in testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
