  • Species reactivity: Human,Mouse
  • Molecular weight guidance: Observed: 90 kDa; Calculated: 52260 MW

Biological background

Protein function (datasheet): Transcriptional modulator activated by BMP (bone morphogenetic proteins) type 1 receptor kinase. SMAD1 is a receptor-regulated SMAD (R-SMAD). SMAD1/OAZ1/PSMB4 complex mediates the degradation of the CREBBP/EP300 repressor SNIP1. May act synergistically with SMAD4 and YY1 in bone morphogenetic protein (BMP)-mediated cardiac-specific gene expression. .

Cellular localization (datasheet): Cytoplasm. Nucleus. Cytoplasmic in the absence of ligand. Migrates to the nucleus when complexed with SMAD4. Co-localizes with LEMD3 at the nucleus inner membrane.

Tissue details (datasheet): Ubiquitous. Highest expression seen in the heart and skeletal muscle.

Research relevance and current trends

  • Commonly studied in contexts related to Growth Factors,Metabolism,Metabolism Processes,Nuclear Signaling,Nuclear Signaling Pathways,Pathways and Processes,Response To Hypoxia,Signal Transduction,Signaling Pathway,Stem Cells,TGF Beta.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a monoclonal antibody, this reagent is expected to recognize a defined epitope, which can support consistency across lots when epitope accessibility is preserved.

We are currently using anti-Smad1 Rabbit Monoclonal antibody M00728-1 for human tissue, and we are happy with the IHC results. The species of reactivity given in the datasheet says human, mouse. Is it possible that the antibody can work on canine tissues as well?
The anti-Smad1 Rabbit Monoclonal antibody (M00728-1) has not been validated for cross reactivity specifically with canine tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in canine you can get your next antibody for free. Please contact me if I can help you with anything.
I am looking for using your anti-Smad1 Rabbit Monoclonal antibody for ub-specific processing proteases studies. Has this antibody been tested with western blotting on hela cell lysate? We would like to see some validation images before ordering.
I appreciate your inquiry. This M00728-1 anti-Smad1 Rabbit Monoclonal antibody is tested on hela cell lysate. It is guaranteed to work for IF, IHC, WB in human, mouse. Our Boster guarantee will cover your intended experiment even if the sample type has not been be directly tested.
