Product Overview
This rabbit polyclonal antibody detects SMAD family member 2 (gene SMAD2) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 52.3 kDa, and the supplier reports an observed band at about 60 kDa.
SMAD2 is a receptor-regulated SMAD that carries signals from TGF-β and activin receptors. After phosphorylation it forms a complex with SMAD4 that enters the nucleus to control gene transcription, so phospho-SMAD2 is a standard readout of TGF-β pathway activity in development, fibrosis and cancer.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human placenta tissue, rat liver tissue, mouse testicular tissue, mouse heart tissue, mouse lung tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human colon cancer tissue, human placenta tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): K562 cells.
Immunogen
Recombinant human SMAD2 fragment (Glu83–Gln264), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places SMAD family member 2 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Expressed at high levels in skeletal muscle, endothelial cells, heart and placenta. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
