Product Overview

This rabbit polyclonal antibody recognizes SMAD family member 4 (gene SMAD4) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 60.4 kDa, and the supplier reports an observed band at about 60 kDa.

SMAD4 is the common partner SMAD that combines with receptor-activated SMADs in both TGF-β and BMP signaling. The resulting complexes enter the nucleus to control gene transcription. SMAD4 is a well-known tumor suppressor, frequently lost in pancreatic and colorectal cancers.

Target SMAD family member 4 (gene SMAD4; UniProt Q13485, human)
Host / Clonality / Isotype Rabbit / Polyclonal / Rabbit IgG
Reactivity Human, Mouse, Rat
Form Lyophilized
Formulation Per vial: 0.9 mg NaCl; 0.2 mg Na2HPO4; 0.05 mg NaN3; plus stabilizers
Calculated MW 60.4 kDa
Observed MW 60 kDa
Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat)

Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue, rat skeletal muscle tissue, mouse skeletal muscle tissue, U87 cells, human placenta tissue, HT1080 cells, HeLa cells, NEURO cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide from the C-terminal region of human smad4. The peptide sequence is conserved in mouse and rat.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places SMAD family member 4 in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on bovine samples?
A.Reactivity with bovine samples has not been tested by the supplier. Aligning the immunogen sequence with the bovine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 60.4 kDa; observed band at about 60 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of human smad4. The peptide sequence is conserved in mouse and rat.
