Product Overview

This rabbit polyclonal antibody recognizes SMAD family member 5 (gene SMAD5) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 52.3 kDa, and the supplier reports an observed band at about 52 kDa.

SMAD5 is a receptor-regulated SMAD in the BMP pathway. When phosphorylated by BMP type I receptors it binds SMAD4 and regulates gene transcription in development, cell differentiation and angiogenesis; the unphosphorylated protein has also been linked to regulation of glycolysis in the cytoplasm.

Validated Applications

IHC (paraffin sections) 0.5–1 µg/mL (Human, Rat, Mouse)
Western blot 0.1–0.5 µg/mL (Human, Rat, Mouse)

Samples with a confirmed band (WB): K562 cells, Jurkat cells, PC 1–2 cells, HeLa cells, SMMC cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): rat intestine tissue, human intestinal cancer tissue, human lung cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide from the middle region of human SMAD5. The peptide sequence is conserved in rat. It differs from the mouse sequence at one residue.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places SMAD family member 5 in the cytoplasm, nucleus and mitochondrion. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Rat, Mouse); IHC (paraffin sections) 0.5–1 µg/mL (Human, Rat, Mouse). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on bovine samples?
A.Reactivity with bovine samples has not been tested by the supplier. Aligning the immunogen sequence with the bovine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 52.3 kDa; observed band at about 52 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the middle region of human SMAD5. The peptide sequence is conserved in rat.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
