Product Overview
This rabbit polyclonal antibody recognizes SMAD family member 9 (gene Smad9) in mouse and rat samples and is validated for Western blot and ELISA. The predicted molecular weight is 48.6 kDa, and the supplier reports an observed band at about 55 kDa.
In mouse, SMAD9 (also called SMAD8) is a receptor-regulated SMAD activated by BMP type I receptors such as ALK2, ALK3 and ALK6. Once phosphorylated it partners with SMAD4 to regulate gene expression, and it may contribute to osteoblast differentiation.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat brain tissue, rat C6 cells, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Recombinant mouse Smad9 fragment (Gln153–His235), expressed in E. coli. Sequence identity with the human and rat orthologs: 59.2% and 96.4%, respectively.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places SMAD family member 9 in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
