Product Overview
This rabbit polyclonal antibody targets guanine nucleotide exchange protein SMCR8 in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 105.0 kDa, and the supplier reports an observed band at about 140 kDa.
SMCR8 forms a complex with C9orf72 and WDR41 that acts as a guanine nucleotide exchange factor for Rab GTPases such as RAB8A and RAB39B. The complex regulates autophagy and lysosome function, and it is studied in the context of C9orf72-linked ALS and frontotemporal dementia.

Samples with a confirmed band (WB): human 293T cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): HEL cells.

Recombinant human SMCR8 fragment (Asp86–Asp776), expressed in E. coli.

The supplier lists reactivity with human. UniProt places guanine nucleotide exchange protein SMCR8 in the cytoplasm, nucleus, presynapse and postsynapse.
