Product Overview

This rabbit polyclonal antibody detects suppressor of cytokine signaling 5 (gene SOCS5) in human, mouse and rat samples and is validated for Western blot, IHC and ELISA. The predicted molecular weight is 61.2 kDa, and the supplier reports an observed band at about 61 kDa.

Suppressor of cytokine signaling 5 (SOCS5) belongs to the SOCS family of negative feedback regulators. It can promote degradation of the EGF receptor, probably as the substrate-recognition subunit of an Elongin BC-cullin ubiquitin ligase, and it restrains IL-4 signaling, which influences the balance of T-helper cell differentiation; it is studied in growth factor and immune signaling.

Validated Applications

Western blot 0.1–0.25 µg/mL (Human, Mouse, Rat)
IHC (paraffin sections) 2–5 µg/mL (Human)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human U87 cells, human U2OS cells, human A549 cells, human K562 cells, human 293T cells, human HeLa cells, rat brain tissue, mouse brain tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human ovarican cancer tissue, human prostatic acinar adenocarcinoma tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Recombinant human SOCS5 fragment (Met386–Lys536), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.25 µg/mL (Human, Mouse, Rat); IHC (paraffin sections) 2–5 µg/mL (Human); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on canine samples?
A.Reactivity with canine samples has not been tested by the supplier. Aligning the immunogen sequence with the canine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 61.2 kDa; observed band at about 61 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant human SOCS5 fragment (Met386–Lys536), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
