Product Overview
Clone 3C4C3 is a mouse monoclonal antibody that recognizes transcription factor Sp1 in human samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 80.7 kDa, and the supplier reports an observed band at about 90 kDa.
Transcription factor Sp1 (SP1) is a zinc finger protein that binds GC-rich promoter elements and regulates a very large number of genes involved in growth, apoptosis, differentiation and immune responses. Its activity is tuned by phosphorylation, acetylation, glycosylation and other modifications, and it is one of the most studied general transcription regulators.

Samples with a confirmed band (WB): human HeLa cells, human Caco-2 cells, human Jurkat cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human colorectal adenocarcinoma tissue, human glioma tissue, human laryngeal squamous cells carcinoma tissue, human lung cancer tissue, human ovarian cancer tissue, human thyroid cancer tissue, human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): A431 cells.

Reactivity Notes
The supplier lists reactivity with human. UniProt places transcription factor Sp1 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Up-regulated in adenocarcinomas of the stomach (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
