Biological background

Protein function (datasheet): Transcription factor that can activate or repress transcription in response to physiological and pathological stimuli. Binds with high affinity to GC-rich motifs and regulates the expression of a large number of genes involved in a variety of processes such as cell growth, apoptosis, differentiation and immune responses. Highly regulated by post-translational modifications (phosphorylations, sumoylation, proteolytic cleavage, glycosylation and acetylation). Binds also the PDGFR- alpha G-box promoter. May have a role in modulating the cellular response to DNA damage. Implicated in chromatin remodeling. Plays a role in the recruitment of SMARCA4/BRG1 on the c-FOS promoter. Plays an essential role in the regulation of FE65 gene expression. In complex with ATF7IP, maintains telomerase activity in cancer cells by inducing TERT and TERC gene expression. Isoform 3 is a stronger activator of transcription than isoform 1. Positively regulates the transcription of the core clock component ARNTL/BMAL1. .

Cellular localization (datasheet): Nucleus. Cytoplasm. Nuclear location is governed by glycosylated/phosphorylated states. Insulin promotes nuclear location, while glucagon favors cytoplasmic location.

Tissue details (datasheet): Up-regulated in adenocarcinomas of the stomach (at protein level). Isoform 3 is ubiquitously expressed at low levels. .

Research relevance and current trends

  • Commonly studied in contexts related to 2339,Domain Families,Epigenetics and Nuclear Signaling,Metabolic Signaling Pathways,Metabolism,Mitochondrial Metabolism,Nuclear,Nucleotide Metabolism,Pathways and Processes,Signaling Pathways,Stem Cells,TGF Beta,Transcription,Zinc Finger.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a monoclonal antibody, this reagent is expected to recognize a defined epitope, which can support consistency across lots when epitope accessibility is preserved.

We have observed staining in human cervix carcinoma. What should we do? Is anti-SP1 Rabbit Monoclonal antibody supposed to stain cervix carcinoma positively?
From what I have seen in literature cervix carcinoma does express SP1. From what I have seen in Uniprot.org, SP1 is expressed in nipple, brain testis, cervix carcinoma, colon, cervix carcinoma erythroleukemia, among other tissues. Regarding which tissues have SP1 expression, here are a few articles citing expression in various tissues: Brain, and Testis, Pubmed ID: 15489334 Cervix carcinoma, Pubmed ID: 10973950, 16964243, 18669648, 20068231 Cervix carcinoma, and Erythroleukemia, Pubmed ID: 23186163 Colon, Pubmed ID: 10976766
