Product Overview
This rabbit polyclonal antibody targets sperm acrosome membrane-associated protein 3 (gene SPACA3) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 23.4 kDa, and the supplier reports an observed band at about 40 kDa.
Sperm acrosome membrane-associated protein 3 (SPACA3, also called SLLP1) is a lysozyme-like protein located in the sperm acrosome. It is thought to take part in sperm-egg adhesion and fusion, possibly by binding N-acetylglucosamine on the egg surface, but it lacks detectable bacteriolytic activity.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human 293T cells, human Daudi cells, rat C6 cells, mouse NIH/3T3 cells, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): Caco-2 cells.
Samples with a confirmed signal (flow cytometry): Daudi cells.
Immunogen
Recombinant human SPACA3 fragment (Glu86–Phe215), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): The processed form is expressed in sperm (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
