Product Overview
Clone 22S18 is a rabbit monoclonal antibody that recognizes speckle-type POZ protein (gene SPOP) in human samples and is validated for Western blot. The predicted molecular weight is 42.1 kDa, and the supplier reports an observed band at about 42 kDa.
Speckle-type POZ protein (SPOP) is the substrate adaptor of a CUL3-based E3 ubiquitin ligase. It targets proteins such as DAXX, GLI2, GLI3 and nuclear receptors and coregulators for ubiquitination, and SPOP is among the most frequently mutated genes in prostate cancer, making it a common subject in cancer research.

Samples with a confirmed band (WB): human Caco-2 cells, human K562 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Reactivity Notes
The supplier lists reactivity with human. UniProt places speckle-type POZ protein in the nucleus, nucleus speckle and cytoplasm. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
