Product Overview

Clone AFGF-19 is a rabbit monoclonal antibody that recognizes spectrin alpha chain, erythrocytic 1 (gene SPTA1) in human samples and is validated for Western blot and IHC. The predicted molecular weight is 280.0 kDa, and the supplier reports an observed band at about 280 kDa.

Spectrin alpha chain, erythrocytic 1 (SPTA1) pairs with β-spectrin to form the flexible network that lines the red blood cell membrane. Together with actin and protein 4.1, this skeleton gives erythrocytes their shape and deformability, and SPTA1 mutations cause hereditary spherocytosis and elliptocytosis.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200

Samples with a confirmed band (WB): K562 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human SPTA1.

Reactivity Notes

The supplier lists reactivity with human. UniProt places spectrin alpha chain, erythrocytic 1 in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on zebrafish samples?
A.Reactivity with zebrafish samples has not been tested by the supplier. Aligning the immunogen sequence with the zebrafish ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 280.0 kDa; observed band at about 280 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human SPTA1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
