Product Overview
This rabbit polyclonal antibody recognizes afadin- and alpha-actinin-binding protein (gene SSX2IP) in human and rat samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 71.2 kDa, and the supplier reports an observed band at about 71 kDa.
SSX2IP (also called ADIP) binds afadin and alpha-actinin and helps organize the actin cytoskeleton at cell–cell adherens junctions. It is also found at the leading edge of migrating cells, where it supports cell movement, and at the centrosome, where it contributes to centrosome maturation and the integrity of the pericentriolar material.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Rat) |
| Flow cytometry (fixed cells) | 1–3 µg /1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human K562 cells, human U251 cells, human A431 cells, human U2OS cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human cervix squamous cells carcinoma tissue, human spleen tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): U251 cells.
Immunogen
Recombinant human SSX2IP fragment (Lys27–Pro614), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 87.9% and 87.4%, respectively.
Reactivity Notes
The supplier lists reactivity with human and rat. UniProt places afadin- and alpha-actinin-binding protein in the cell junction and nucleus. Tissue expression noted by UniProt (human): Widely expressed, with the highest expression in brain, intermediate expression in kidney, testis, spinal cord, liver, heart, lung, skeletal muscle, ovary, fetal liver and fetal brain, and little to no expression in pancreas and spleen. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
