Overview

This antibody is intended for detection of STAT1 (Signal transducer and activator of transcription 1-alpha/beta) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-STAT1 Antibody Picoband® catalog # PB9510. Tested in Flow Cytometry, WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the N-terminus of human STAT1, different from the related mouse sequence by two amino acids.
  • Molecular weight context: reported MW: 83, 91 kDa; calculated MW: 83,043 MW, 87335 MW
  • Reactivity: Human,Mouse,Rat
  • Applications: Flow Cytometry, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Signal transducer and activator of transcription 1-alpha/beta; signal transducer and activator of transcription 1, 91kDa. Signal transducer and activator of transcription 1 (STAT1) is a transcription factor which in humans is encoded by the STAT1 gene. The protein encoded by this gene is a member of the STAT protein family. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein can be activated by various ligands including interferon-alpha, interferon-gamma, EGF, PDGF and IL6. This protein mediates the expression of a variety of genes, which is thought to be important for cell viability in response to different cell stimuli and pathogens. Two alternatively spliced transcript variants encoding distinct isoforms have been described. Functional note: Signal transducer and transcription activator that mediates cellular responses to interferons (IFNs), cytokine KITLG/SCF and other cytokines and other growth factors. Following type I IFN (IFN-alpha and IFN-beta) binding to cell surface receptors, signaling via protein kinases leads to activation of Jak kinases (TYK2 and JAK1) and to tyrosine phosphorylation of STAT1 and STAT2. The phosphorylated STATs dimerize and associate with ISGF3G/IRF-9 to form a complex termed ISGF3 transcription factor, that enters the nucleus. ISGF3 binds to the IFN stimulated response element (ISRE) to activate the transcription of IFN- stimulated genes (ISG), which drive the cell in an antiviral state. In response to type II IFN (IFN-gamma), STAT1 is tyrosine- and serine-phosphorylated. It then forms a homodimer termed IFN- gamma-activated factor (GAF), migrates into the nucleus and binds to the IFN gamma activated sequence (GAS) to drive the expression of the target genes, inducing a cellular antiviral state. Becomes activated in response to KITLG/SCF and KIT signaling. May mediate cellular responses to activated FGFR1, FGFR2, FGFR3 and FGFR4. . Reported localization: Cytoplasm . Nucleus . Translocated into the nucleus upon tyrosine phosphorylation and dimerization, in response to IFN-gamma and signaling by activated FGFR1, FGFR2, FGFR3 or FGFR4. Expression/tissue context: Ubiquitously expressed.

Research relevance and current trends

  • Cancer: Researchers commonly examine how STAT1 (Signal transducer and activator of transcription 1-alpha/beta) relates to this theme using model systems and orthogonal readouts.

  • Nuclear Signaling: Researchers commonly examine how STAT1 (Signal transducer and activator of transcription 1-alpha/beta) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative STAT1 (Signal transducer and activator of transcription 1-alpha/beta) levels across conditions; band patterns may reflect isoforms and processing.
  • Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.

Notes for experimental interpretation

  • Family / similarity context: Belongs to the transcription factor STAT family.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

My colleagues were satisfied with the WB result of your anti-STAT1 antibody. However we have observed positive staining in placenta testis cytoplasm using this antibody. Is that expected? Could you tell me where is STAT1 supposed to be expressed?
From what I have seen in literature, placenta testis does express STAT1. Generally STAT1 expresses in cytoplasm. Regarding which tissues have STAT1 expression, here are a few articles citing expression in various tissues: B-cell, Pubmed ID: 9724754 Brain, Pubmed ID: 15489334 Cervix carcinoma, Pubmed ID: 18669648, 18691976, 20068231 Cervix carcinoma, and Erythroleukemia, Pubmed ID: 23186163 Leukemic T-cell, Pubmed ID: 19690332 Liver, Pubmed ID: 24275569 Placenta, and Testis, Pubmed ID: 14702039
