Product Overview
This rabbit polyclonal antibody targets serine/threonine-protein kinase 16 (gene STK16) in human, mouse and rat samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 34.7 kDa, and the supplier reports an observed band at about 37 kDa.
STK16 (also called PKL12 or MPSK1) is a membrane-associated serine/threonine kinase found at the Golgi. It can autophosphorylate and phosphorylate substrates such as DRG1 and 4E-BP1 in vitro, and it has been linked to secretory vesicle trafficking and TGF-β signaling, though its physiological roles are not fully defined.

|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Mouse, Rat) |
| Flow cytometry (fixed cells) | 1–3 µg /1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human THP-1 cells, human Jurkat cells, human HEL cells, human HepG2 cells, rat liver tissue, rat kidney tissue, mouse liver tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse testis tissue, rat testis tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): RT4 cells.
Immunogen
Recombinant human STK16 fragment (Glu109–Leu268), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 95.6% and 96.9%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places serine/threonine-protein kinase 16 in the cytoplasm and membrane. Tissue expression noted by UniProt (human): Ubiquitously expressed at very low levels. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
