Product Overview

Clone 25S37 is a rabbit monoclonal antibody that recognizes syntaxin-1A (gene STX1A) in human, mouse and rat samples and is validated for Western blot, immunoprecipitation and flow cytometry. The predicted molecular weight is 33.0 kDa, and the supplier reports an observed band at about 33 kDa.

Syntaxin-1A (STX1A) is a plasma membrane SNARE that forms the core fusion complex with SNAP25 and the vesicle SNARE VAMP2. Zippering of these SNAREs brings synaptic vesicles and the presynaptic membrane together for calcium-triggered neurotransmitter release, and STX1A also supports hormone secretion from endocrine cells.

Validated Applications

Western blot 1:1000–1:5000
Immunoprecipitation 1:50
Flow cytometry 1:50

Samples with a confirmed band (WB): SH-SY5Y cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human STX1A.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places syntaxin-1A in the synapse and cell membrane. Tissue expression noted by UniProt (human): Highly expressed in embryonic spinal cord and ganglia and in adult cerebellum and cerebral cortex... The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:1000–1:5000; Flow cytometry 1:50; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 33.0 kDa; observed band at about 33 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human STX1A.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
