Product Overview
This rabbit polyclonal antibody recognizes copper chaperone for superoxide dismutase (gene CCS) in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 29.0 kDa, and the supplier reports an observed band at about 29 kDa.
CCS (copper chaperone for superoxide dismutase) delivers copper ions to Cu/Zn superoxide dismutase (SOD1) and helps form its disulfide bond, both steps needed for SOD1 activity. It is studied in copper homeostasis, oxidative stress and the biology of SOD1 in neurodegeneration.

Validated Applications
| IHC (paraffin sections) | 0.5–1 µg/mL (Human) |
|---|---|
| Western blot | 0.1–0.5 µg/mL (Human, Mouse, Rat) |
Samples with a confirmed band (WB): human A549 cells, rat heart tissue, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Immunogen
Synthetic peptide from the C-terminal region of human CCS. It differs from the mouse sequence at six residues and from the rat sequence at seven residues.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places copper chaperone for superoxide dismutase in the cytoplasm. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
