Product Overview
This rabbit polyclonal antibody recognizes tyrosine-protein kinase SYK in human, mouse and rat samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 72.1 kDa, and the supplier reports an observed band at about 72 kDa.
Spleen tyrosine kinase (SYK) is a non-receptor tyrosine kinase that binds phosphorylated ITAM motifs in immunoreceptors through its tandem SH2 domains. It relays signals from B-cell receptors, Fc receptors, C-type lectin receptors and platelet receptors, driving immune activation, phagocytosis and platelet function, and it is a target in autoimmune and B-cell cancer research.

Samples with a confirmed band (WB): human THP-1 cells, human HEL cells, human A431 cells, human RT4 cells, rat spleen tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lymphoma tissue, mouse spleen tissue, rat spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): RT4 cells.

Recombinant human SYK fragment (Ala10–Asn635), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places tyrosine-protein kinase SYK in the cell membrane and cytoplasm. Tissue expression noted by UniProt (human): Widely expressed in hematopoietic cells (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
