Biological background

CD81 antigen. CD81 molecule, also known as CD81 (Cluster of Differentiation 81), is a protein which in humans is encoded by the CD81 gene. The protein encoded by this gene is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is a cell surface glycoprotein that is known to complex with integrins. This protein appears to promote muscle cell fusion and support myotube maintenance. Also it may be involved in signal transduction. This gene is localized in the tumor-suppressor gene region and thus it is a candidate gene for malignancies. Two transcript variants encoding different isoforms have been found for this gene. Functional note: May play an important role in the regulation of lymphoma cell growth. May be involved in the acrosome reaction. Reported localization: Basolateral cell membrane. Expression/tissue context: Expressed in oocytes. Highly expressed in granulosa cells.

We are currently using anti-TAPA1/Cd81 antibody A01281-3 for rat tissue, and we are well pleased with the IHC results. The species of reactivity given in the datasheet says mouse, rat. Is it possible that the antibody can work on pig tissues as well?
The anti-TAPA1/Cd81 antibody (A01281-3) has not been validated for cross reactivity specifically with pig tissues, though there is a good chance of cross reactivity. Would A01281-3 anti-TAPA1/Cd81 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
You can see on the product datasheet, A01281-3 anti-TAPA1/Cd81 antibody as been tested on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
