Product Overview

This rabbit polyclonal antibody detects C-C motif chemokine 17 (gene Ccl17) in mouse samples and is validated for Western blot and ELISA.

CCL17 (also called TARC) is a C-C chemokine that binds the receptor CCR4 and preferentially attracts Th2 cells and regulatory T cells. It is produced by dendritic cells and other cells and is elevated in allergic diseases such as atopic dermatitis, where it is commonly measured as a disease activity marker.

Target C-C motif chemokine 17 (gene Ccl17; UniProt Q9WUZ6, mouse)
Host / Clonality / Isotype Rabbit / Polyclonal / Rabbit IgG
Reactivity Mouse
Form Lyophilized
Formulation Per vial: 0.9 mg NaCl; 0.2 mg Na2HPO4; 0.05 mg NaN3; plus stabilizers
Observed MW 34 kDa
Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

Western blot 0.1–0.5 µg/mL (Mouse)
ELISA 0.1–0.5 µg/mL

Conditions in the example images (WB): 5–20% gradient SDS-PAGE, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Immunogen

Recombinant mouse TARC fragment (Ala34–Pro103), expressed in E. coli. Sequence identity with the human ortholog: 71%.

Reactivity Notes

The supplier lists reactivity with mouse. UniProt describes C-C motif chemokine 17 as a secreted protein. Tissue expression noted by UniProt (mouse): Expressed in primary and secondary lymphoid organs, where it is specifically produced by a subset of dendritic cells.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Mouse); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Mouse. Other species are not listed by the supplier.
Q.Has this antibody been tested on canine samples?
A.Reactivity with canine samples has not been tested by the supplier. Aligning the immunogen sequence with the canine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: observed band at about 34 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant mouse TARC fragment (Ala34–Pro103), expressed in E. coli. Sequence identity with the human ortholog: 71%.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
