Product Overview

Clone ACOI-20 is a rabbit monoclonal antibody that recognizes TATA box-binding protein-like 1 (gene TBPL1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 20.9 kDa, and the supplier reports an observed band at about 23 kDa.

TBP-like protein 1 (TBPL1, also called TRF2) is a relative of TATA-box-binding protein that does not bind the TATA box. It supports a specialized transcription system at promoters carrying the TCT core element, which drives expression of most ribosomal protein genes.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:20
Flow cytometry 1:20

Samples with a confirmed band (WB): HeLa cells, mouse testis.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human TBPL1 Does not bind the TATA box.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places TATA box-binding protein-like 1 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Ubiquitously expressed, with highest levels in the testis and ovary. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:20; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on monkey samples?
A.Reactivity with monkey samples has not been tested by the supplier. Aligning the immunogen sequence with the monkey ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 20.9 kDa; observed band at about 23 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human TBPL1 Does not bind the TATA box.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
