Product Overview
This rabbit polyclonal antibody detects TAR DNA-binding protein 43 (gene TARDBP) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry, immunoprecipitation and ELISA. The predicted molecular weight is 44.7 kDa, and the supplier reports an observed band at about 43 kDa.
TDP-43 (TARDBP) is a nuclear RNA-binding protein with two RRM domains that binds GU-rich sequences to regulate splicing, mRNA stability and transport. Cytoplasmic aggregates of TDP-43 are a pathological hallmark of most ALS cases and of frontotemporal lobar degeneration, making it a central protein in neurodegeneration research.

Samples with a confirmed band (WB): human HeLa cells, human Jurkat cells, human HepG2 cells, rat C6 cells, mouse pancrease, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colorectal adenocarcinoma tissue, human liver cancer tissue, human ovarian cancer tissue, human spleen tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed result (IP): HepG2 cells.
Samples with a confirmed signal (flow cytometry): Jurkat cells.
Immunogen
Recombinant human TDP-43/TARDBP fragment (Met1–His264), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places TAR DNA-binding protein 43 in the nucleus, cytoplasm and mitochondrion. Tissue expression noted by UniProt (human): Ubiquitously expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
