| Immunogen | A synthesized peptide derived from human TDT Template-independent DNA polymerase which catalyzes the random addition of deoxynucleoside 5'-triphosphate to the 3'-end of a DNA initiator. One of the in vivo functions of this enzyme is the addition of nucleotides at the junction (N region) of rearranged Ig heavy chain and T-cell receptor gene segments during the maturation of B- and T-cells. |

Product Overview
Clone AFHA-4 is a rabbit monoclonal antibody that recognizes DNA nucleotidylexotransferase (gene DNTT) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 58.5 kDa, and the supplier reports an observed band at about 59 kDa.
Terminal deoxynucleotidyl transferase (TdT, encoded by DNTT) is a DNA polymerase that adds nucleotides to DNA ends without a template. In developing B and T cells, it inserts random N nucleotides at V(D)J junctions, increasing the diversity of antibodies and T-cell receptors. TdT is a classic marker of immature lymphoid cells and lymphoblastic leukemia.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunohistochemistry | 1:50–1:200 |
Samples with a confirmed band (WB): human Jurkat cells, human MOLT-4 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human TDT Template-independent DNA polymerase which catalyzes the random addition of deoxynucleoside 5'-triphosphate to the 3'-end of a DNA initiator.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places DNA nucleotidylexotransferase in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
