Product Overview

This rabbit polyclonal antibody recognizes tissue factor pathway inhibitor 2 (gene TFPI2) in human samples and is validated for Western blot and IHC. The predicted molecular weight is 26.9 kDa, and the supplier reports an observed band at about 27–30 kDa.

Tissue factor pathway inhibitor 2 (TFPI2) is a Kunitz-type serine protease inhibitor secreted into the extracellular matrix. It inhibits plasmin, trypsin and the factor VIIa–tissue factor complex, and it is studied in matrix remodeling, placental biology and as a gene often silenced by methylation in cancers.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human)
IHC (paraffin sections) 2–5 µg/mL (Human)

Samples with a confirmed band (WB): human placenta tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human placenta tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide from the middle region of human TFPI2.

Reactivity Notes

The supplier lists reactivity with human. UniProt describes tissue factor pathway inhibitor 2 as a secreted protein. Tissue expression noted by UniProt (human): Umbilical vein endothelial cells, liver, placenta, heart, pancreas, and maternal serum at advanced pregnancy. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human); IHC (paraffin sections) 2–5 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on primate samples?
A.Reactivity with primate samples has not been tested by the supplier. Aligning the immunogen sequence with the primate ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 26.9 kDa; observed bands at about 27 and 30 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the middle region of human TFPI2.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
