Product Overview

Clone AFEC-20 is a rabbit monoclonal antibody that recognizes thyroglobulin (gene TG) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 304.8 kDa, and the supplier reports an observed band at about 305 kDa.

Thyroglobulin (TG) is a very large glycoprotein made by thyroid follicular cells and stored in the follicle lumen. Selected tyrosines on thyroglobulin are iodinated and coupled to form the thyroid hormones T4 and T3, which are released after thyroglobulin is taken back up and degraded; serum thyroglobulin is also a standard marker in thyroid cancer follow-up.

Immunocytochemistry / Immunofluorescence 1:50–1:200
Flow cytometry 1:50

Samples with a confirmed band (WB): mouse thyroid.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human thyroid cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human thyroglobulin.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt describes thyroglobulin as a secreted protein. Tissue expression noted by UniProt (human): Specifically expressed in the thyroid gland. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
