Product Overview
This rabbit polyclonal antibody detects angiopoietin-1 receptor (gene TEK) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 125.8 kDa, and the supplier reports an observed band at about 160 kDa.
TIE2 (TEK) is a receptor tyrosine kinase on endothelial cells that binds angiopoietin-1 and angiopoietin-2. Angiopoietin-1 signaling through TIE2 promotes endothelial survival and vascular stability, while angiopoietin-2 can oppose it, making TIE2 a central subject in angiogenesis, vascular leak and lymphatic development research.

Samples with a confirmed band (WB): human placenta tissue, human Caco-2 cells, human HeLa cells, rat liver tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue, human mammary cancer tissue, human placenta tissue, mouse heart tissue, mouse lung tissue, rat heart tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Immunogen
Synthetic peptide from the N-terminal region of human TIE2/TEK. The peptide sequence is conserved in mouse and rat.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes angiopoietin-1 receptor as secreted and also located in the cell membrane, cell junction and cytoplasm. Tissue expression noted by UniProt (human): Detected in umbilical vein endothelial cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
