Product Overview
This rabbit polyclonal antibody recognizes angiopoietin-1 receptor (gene TEK) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 125.8 kDa, and the supplier reports an observed band at about 160 kDa.
TIE2 (TEK) is a receptor tyrosine kinase on endothelial cells that binds angiopoietin-1 and angiopoietin-2. Angiopoietin-1 signaling through TIE2 promotes endothelial survival and vascular stability, while angiopoietin-2 can oppose it, making TIE2 a central subject in angiogenesis, vascular leak and lymphatic development research.

Samples with a confirmed band (WB): mouse lung tissue, mouse Neuro-2a cells, human HeLa cells, human U-87MG cells, human U2OS cells, human A431 cells, human PC-3 cells, human HL-60 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): rat lung tissue, human tonsil tissue, human ovarian cancer tissue, rat intestine tissue, mouse testis tissue, rat testis tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): HeLa cells.

Recombinant human TIE2/TEK fragment (Ala23–Arg616), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes angiopoietin-1 receptor as secreted and also located in the cell membrane, cell junction and cytoplasm. Tissue expression noted by UniProt (human): Detected in umbilical vein endothelial cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
