Product Overview
This rabbit polyclonal antibody detects T-cell immunoglobulin and mucin domain-containing protein 2 (gene Timd2) in mouse and rat samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 33.5 kDa, and the supplier reports an observed band at about 70 kDa.
In mouse, TIM-2 (Timd2) is a cell surface glycoprotein of the TIM family that acts as a receptor for H-ferritin. It mediates ferritin uptake by endocytosis in tissues such as liver and kidney, contributing to iron handling, and it has also been linked to regulation of T-cell responses. TIM-2 is studied in iron homeostasis and immunology.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Mouse, Rat) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Mouse) |
Samples with a confirmed band (WB): mouse liver tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse liver tissue, rat liver tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): HEPA1–6 cells.
Immunogen
Synthetic peptide from the C-terminal region of mouse tim 2/Timd2. Sequence identity with the rat ortholog: 73.9%.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places T-cell immunoglobulin and mucin domain-containing protein 2 in the cell membrane. Tissue expression noted by UniProt (mouse): Expressed on late differentiated Th2 cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
