Product Overview
Clone 22T48 is a rabbit monoclonal antibody that recognizes protein timeless homolog in human samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 138.7 kDa, and the supplier reports an observed band at about 150 kDa.
TIMELESS is a replication fork protection factor that forms a complex with TIPIN at the replisome. The complex couples helicase and polymerase activities, stabilizes stalled forks and supports checkpoint signaling to protect genome stability. TIMELESS also has links to the circadian clock and is studied in DNA replication stress, genome maintenance and cancer.

Samples with a confirmed band (WB): human HeLa cells, human A549 cells, human MCF-7 cells, human K562 cells, human SIHA cells, human RT4 cells, human Jurkat cells, human HEL cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

The supplier lists reactivity with human. UniProt places protein timeless homolog in the nucleus and chromosome. Tissue expression noted by UniProt (human): Expressed in all tissues examined including brain, heart, lung, liver, skeletal muscle, kidney, placenta, pancreas, spleen, thymus and testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
