Product Overview

Clone GGE-6 is a rabbit monoclonal antibody that recognizes tissue factor (gene F3) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 33.1 kDa, and the supplier reports an observed band at about 50 kDa.

Tissue factor (F3, CD142) is a transmembrane glycoprotein that starts the extrinsic pathway of blood coagulation. When exposed to blood it binds factor VII/VIIa, and the complex activates factors IX and X. Tissue factor is expressed by cells outside the vessel wall and can be induced on monocytes and tumor cells, linking coagulation with inflammation and cancer.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human SH-SY5Y cells, human A549 cells, human PANC-1 cells, rat lung tissue, rat liver tissue, mouse lung tissue, mouse liver tissue lysates.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Samples with confirmed staining (IHC): human colon cancer tissue, human lung cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human tissue Factor.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Lung, placenta and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 33.1 kDa; observed band at about 50 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human tissue Factor.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
