Biological background

Protein function (datasheet): Cooperates with LY96 to mediate the innate immune response to bacterial lipoproteins and other microbial cell wall components. Cooperates with TLR1 or TLR6 to mediate the innate immune response to bacterial lipoproteins or lipopeptides. Acts via MYD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. May also promote apoptosis in response to lipoproteins. Recognizes mycoplasmal macrophage-activating lipopeptide-2kD (MALP-2), soluble tuberculosis factor (STF), phenol-soluble modulin (PSM) and B.burgdorferi outer surface protein A lipoprotein (OspA-L) cooperatively with TLR6.

Cellular localization (datasheet): Membrane ; Single-pass type I membrane protein .

Tissue details (datasheet): Highly expressed in peripheral blood leukocytes, in particular in monocytes, in bone marrow, lymph node and in spleen. Also detected in lung and in fetal liver. Levels are low in other tissues.

Research relevance and current trends

  • Commonly studied in contexts related to Adapters,Atherosclerosis,Cardiovascular,Defensin,Human Protein,Immunoglobulins,Immunology,Innate Immunity,Macrophage/Inflammation,Microbiology,Protein,Receptors,Signal Transduction,TLR Signaling,Transmembrane,Vascular Inflammation.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a monoclonal antibody, this reagent is expected to recognize a defined epitope, which can support consistency across lots when epitope accessibility is preserved.

My team were content with the WB result of your anti-TLR2 Rabbit Monoclonal antibody. However we have seen positive staining in fetal lung membrane using this antibody. Is that expected? Could you tell me where is TLR2 supposed to be expressed?
From literature, fetal lung does express TLR2. Generally TLR2 expresses in membrane. Regarding which tissues have TLR2 expression, here are a few articles citing expression in various tissues: Blood, Pubmed ID: 15489334 Fetal lung, Pubmed ID: 9751057 Leukocyte, and Prostate, Pubmed ID: 9596645, 9435236 Monocyte, Pubmed ID: 16622205 T-cell, Pubmed ID: 10426995, 21078852
Can you help my question with product M00131, anti-TLR2 Rabbit Monoclonal antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?
We do not advise storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free M00131 anti-TLR2 Rabbit Monoclonal antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
