Overview

This antibody is intended for detection of TLR5 (Toll-like receptor 5) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-TLR5 Antibody Picoband® catalog # PB9451. Tested in Flow Cytometry, IHC, ICC, WB applications. This antibody reacts with Human. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of human TLR5, different from the related mouse sequence by five amino acids.
  • Molecular weight context: reported MW: 98 kDa; calculated MW: 97834 MW
  • Reactivity: Human
  • Applications: Flow Cytometry, IHC, ICC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Toll-like receptor 5; Toll-like receptor 5. TLR5 encodes a member of the toll-like receptor (TLR) family, which plays a fundamental role in pathogen recognition and activation of innate immune responses. These receptors recognize distinct pathogen-associated molecular patterns that are expressed on infectious agents. The protein encoded by this gene recognizes bacterial flagellin, the principal component of bacterial flagella and a virulence factor. The activation of this receptor mobilizes the nuclear factor NF-kappaB, which in turn activates a host of inflammatory-related target genes. Mutations in this gene have been associated with both resistance and susceptibility to systemic lupus erythematosus, and susceptibility to Legionnaire disease. Functional note: Participates in the innate immune response to microbial agents. Mediates detection of bacterial flagellins. Acts via MYD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. . Reported localization: Membrane ; Single-pass type I membrane protein . Expression/tissue context: Highly expressed in ovary and in peripheral blood leukocytes, especially in monocytes, less in CD11c+ immature dendritic cells. Also detected in prostate and testis.

Research relevance and current trends

  • Immunoglobulins: Researchers commonly examine how TLR5 (Toll-like receptor 5) relates to this theme using model systems and orthogonal readouts.
  • Immunology: Researchers commonly examine how TLR5 (Toll-like receptor 5) relates to this theme using model systems and orthogonal readouts.
  • Innate Immunity: Researchers commonly examine how TLR5 (Toll-like receptor 5) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative TLR5 (Toll-like receptor 5) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of TLR5 (Toll-like receptor 5) across tissue regions and cell types using matched controls.
  • IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
  • Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Family / similarity context: Belongs to the Toll-like receptor family.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

Would PB9451 anti-TLR5 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
It shows on the product datasheet, PB9451 anti-TLR5 antibody as been tested on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
