Product Overview
This rabbit polyclonal antibody detects toll-like receptor 8 (gene Tlr8) in mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 119.4 kDa, and the supplier reports an observed band at about 119 kDa.
Toll-like receptor 8 (TLR8) is an endosomal pattern recognition receptor that recognizes fragments of single-stranded RNA, particularly GU-rich sequences from viruses and bacteria. It signals through MyD88 to activate NF-κB and pro-inflammatory cytokines, mainly in monocytes and myeloid dendritic cells. TLR8 is studied in antiviral immunity and as a target for vaccine adjuvants.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Mouse, Rat) |
Samples with a confirmed band (WB): mouse Ana-1 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse lung tissue, rat lung tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the N-terminal region of mouse tlr8. Sequence identity with the human ortholog: 76.5%.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places toll-like receptor 8 in the endosome membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
