Product Overview
This rabbit polyclonal antibody targets RNA-binding protein FUS in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 53.4 kDa, and the supplier reports an observed band at about 70 kDa.
FUS is a nuclear DNA/RNA-binding protein of the FET family that takes part in transcription, splicing, mRNA transport and the DNA damage response. It has a low-complexity domain that can drive liquid–liquid phase separation. Mutations in FUS cause some forms of amyotrophic lateral sclerosis (ALS), and FUS inclusions are seen in a subset of frontotemporal lobar degeneration.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse, Rat) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human, Mouse) |
Samples with a confirmed band (WB): human HeLa cells, human K562 cells, human 293T cells, human Caco-2 cells, human Jurkat cells, human HL-60 cells, human HepG2 cells, human SH-SY5Y cells, rat brain tissue, rat PC-12 cells, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human rectal cancer tissue, mouse colon tissue, rat colon tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with a confirmed signal (flow cytometry): ANA-1 cells, U87 cells.
Immunogen
Synthetic peptide from the middle region of human TLS/FUS. The peptide sequence is conserved in mouse.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places RNA-binding protein FUS in the nucleus. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
