Product Overview
This rabbit polyclonal antibody recognizes E3 ubiquitin-protein ligase TM129 (gene TMEM129) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 40.5 kDa, and the supplier reports an observed band at about 50 kDa.
TMEM129 is an endoplasmic reticulum membrane E3 ubiquitin ligase that works with the E2 enzyme UBE2J2 in ER-associated degradation. It is hijacked by the human cytomegalovirus protein US11 to degrade HLA class I heavy chains, helping the virus evade immune detection. TMEM129 is studied in protein quality control and viral immune evasion.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human T-47D cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): SiHa cells.
Immunogen
Recombinant human TMEM129 fragment (Glu25–Arg362), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places E3 ubiquitin-protein ligase TM129 in the endoplasmic reticulum membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
