Product Overview
This rabbit polyclonal antibody detects transmembrane protein 59 (gene TMEM59) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 36.2 kDa, and the supplier reports an observed band at about 44 kDa.
TMEM59 is a type I transmembrane protein found in late endosomes and lysosomes. Through a short motif that binds ATG16L1, it promotes LC3 lipidation on the membranes where it resides, triggering a local form of autophagy that targets bacteria such as Staphylococcus aureus. TMEM59 is studied in autophagy, host defense and membrane trafficking.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human MCF-7 cells, human SiHa cells, human U-87MG cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): RT4 cells.
Immunogen
Recombinant human TMEM59 fragment (Ala37–Ile323), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places transmembrane protein 59 in the late endosome membrane, lysosome membrane, cell membrane and Golgi apparatus membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
