Product Overview
This rabbit polyclonal antibody targets transmembrane protein 65 (gene TMEM65) in human, mouse and rat samples and is validated for Western blot and flow cytometry. The predicted molecular weight is 25.5 kDa, and the supplier reports an observed band at about 20 kDa.
TMEM65 is a mitochondrial inner membrane protein that supports sodium-dependent calcium efflux from mitochondria. It associates with the Na+/Ca2+ exchanger NCLX (SLC8B1) and appears to promote its activity, although its exact mechanism is still unclear. TMEM65 is studied in mitochondrial calcium handling and in heart and muscle physiology.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human, Mouse, Rat) |
Samples with a confirmed band (WB): human HeLa cells, human PC-3 cells, human 293T cells, human MCF-7 cells, rat brain tissue, rat C6 cells, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): SiHa cells, ANA-1 cells, RH35 cells.
Immunogen
Synthetic peptide from the middle region of human TMEM65. The peptide sequence is conserved in mouse.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places transmembrane protein 65 in the mitochondrion inner membrane and cell membrane. Tissue expression noted by UniProt (human): Predominantly expressed the ventricular tissue (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
