Product Overview
This rabbit polyclonal antibody detects tumor necrosis factor (gene Tnf) in mouse samples and is validated for Western blot. The predicted molecular weight is 25.9 kDa, and the supplier reports an observed band at about 25 kDa.
Tumor necrosis factor (TNF, TNF-α) is a pro-inflammatory cytokine produced mainly by activated macrophages and T cells. Acting through the receptors TNFR1 and TNFR2, it activates NF-κB and MAP kinase pathways and can trigger apoptosis or necroptosis, driving fever and inflammation. TNF is a central target in rheumatoid arthritis and inflammatory bowel disease research and a standard readout in immunology studies.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Mouse) |
|---|
Samples with a confirmed band (WB): mouse thymus tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Immunogen
Synthetic peptide from the C-terminal region of mouse TNF alpha. It differs from the human sequence at five residues and from the rat sequence at three residues.
Reactivity Notes
The supplier lists reactivity with mouse. UniProt places tumor necrosis factor in the cell membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
