Biological background

Protein function (datasheet): Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation.

Scientific background (datasheet): TNF alpha (Tumor Necrosis Factor alpha) gene encodes a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. This cytokine is mainly secreted by macrophages. It can bind to, and thus functions through its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. This cytokine is involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. This cytokine has been implicated in a variety of diseases, including autoimmune diseases, insulin resistance, and cancer. Knockout studies in mice also suggested the neuroprotective function of this cytokine.

Cellular localization (datasheet): Cell membrane; Single-pass type II membrane protein.

Tissue details (datasheet): Found in spleen (B-cell-rich zone or follicles), Peyer patches (strongest within germinal centers and extending to the mantle zone) and lymph nodes (in reticular pattern in follicles).

Research relevance and current trends

  • Commonly studied in contexts related to TNF Superfamily,Vascular Inflammation.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Cross-reactivity (datasheet): No cross-reactivity with other proteins
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.

We have seen staining in mouse prostatic carcinoma. Any tips? Is anti-TNF alpha antibody supposed to stain prostatic carcinoma positively?
From literature prostatic carcinoma does express TNF. From Uniprot.org, TNF is expressed in leukocyte, blood, prostatic carcinoma, among other tissues. Regarding which tissues have TNF expression, here are a few articles citing expression in various tissues: Blood, Pubmed ID: 15489334 Prostatic carcinoma, Pubmed ID: 8597870, 10205166
Is this RP1000 anti-TNF alpha antibody reactive to the isotypes of TNF?
The immunogen of RP1000 anti-TNF alpha antibody is E. coli-derived mouse TNF alpha recombinant protein(Position: L80-L235). Could you tell me which isotype you are interested in so I can help see if the immunogen is part of this isotype?
