Product Overview
This rabbit polyclonal antibody detects tumor necrosis factor ligand superfamily member 18 (gene TNFSF18) in human samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 22.7 kDa, and the supplier reports an observed band at about 15 kDa.
GITR ligand (GITRL, TNFSF18) is a TNF family cytokine that binds the receptor GITR (TNFRSF18). It costimulates T cells by lowering their activation threshold, activates NF-κB, and increases endothelial VCAM-1 and ICAM-1 expression to support leukocyte adhesion. GITRL is studied in T-cell regulation, regulatory T-cell biology and inflammation.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL |
| ELISA (capture antibody) | 1–5 µg/mL |
Conditions in the example images (WB): 5–20% gradient SDS-PAGE, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human colon cancer tissue, human lung cancer tissue, human mammary cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Immunogen
Recombinant human TNFSF18 fragment (Phe48–Ser177), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places tumor necrosis factor ligand superfamily member 18 in the cell membrane. Tissue expression noted by UniProt (human): Expressed at high levels in the small intestine, ovary, testis, kidney and endothelial cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
